تاثیر متیل جاسمونات و هیدروژن پراکسید بر میزان تاکسول در کشت سوسپانسون سلولی فندق

چکیده

کشت سوسپانسیون سلولی فندق به عنوان روشی مؤثر برای تولید متابولیت‌های ثانویه و افزایش بهره‌وری در تحقیقات بیوشیمیایی و زیست‌فناوری به کار می‌رود. این فناوری امکان کنترل دقیق شرایط رشد، کاهش هزینه‌ها و تولید انبوه ترکیبات دارویی مانند تاکسول را فراهم می‌کند. پژوهش حاضر به منظور بررسی اثرات متیل جاسمونات و پراکسید هیدروژن (H2O2) یر میزان متابولیت‌های ثانویه فندق طی کشت سوسپانسیون سلولی فندق انجام شد. تیمارها شامل متیل جاسمونات در چهار غلظت 100، 150، 250 و 350 میلی گرم در لیتر و آب اکسیژنه در غلطت 0/5 و 1 میلی لیتر در لیتر همراه با تیمار شاهد بود. بعد از اتمام آزمایش صفاتی مانند وزن تر، وزن خشک، محتوای فنل و فلاونوئید کل و تاکسول انجام شد. نتایج نشان داد که استفاده از متیل جاسمونات و H2O2 بر میزان متابولیت‌های مورد بررسی در سوسپانسیون سلولی فندق تاثیر چشمگیری داشت. استفاده از غلظت 150 و 250 میلی گرم در لیتر متیل جاسمونات نسبت به غلظت‌های مختلف پراکسید هیدروژن تاثیر بهتری بر میزان محتوای فنل و فلاونوئید کل داشت. میزان محتوای فنل (14/88 میلی‌گرم در گرم وزن خشک) و فلاونوئید کل (2/65 میلی‌گرم در گرم وزن خشک) در نمونه‌های سوسپانسیون سلولی تیمار شده با متیل جاسمونات در غلظت 250 میلی گرم در لیتر مشاهده شد. همچنین نتایج نشان داد که بیشترین میزان تاکسول ( 308/79 میلی گرم در لیتر) در نمونه‌های تیمار شده با پراکسید هیدروژن در غلظت 0/5 میلی لیتر مشاهده شد.
